研究成果

阿尔茨海默病(Alzheimer’s disease, AD)早期的发病特点、病理特征及演变规律已成为AD早期诊断和干预治疗的主流方向之一。郭腾飞博士牵头建立大湾区老年脑健康计划(Greater-Bay-Area Healthy Aging Brain Study, GHABS)衰老队列(ClinicalTrials注册号:NCTO6183658,Alzheimer’s Research & Therapy2024),瞄准AD这一重大科学难题与国际发展前沿,结合痴呆相关的神经认知评分、生物化学技术、多模态神经影像等领域内的高新技术手段,揭示粤港澳大湾区老年人群的AD病理特征、演变规律、风险因素及保护指标,开发AD早期诊断新技术和早期干预新靶点,为我国AD早诊优治提供科学依据。

主要学术成果包括:①按照AD队列国际标准,牵头建立“大湾区老年脑健康计划”AD社区队列,首次报道了深圳及周边城市社区中老年人的AD发病情况,并基于此揭示了血浆生物标记物和多模态神经影像在我国AD病人发生发展中的演变规律(Nature Aging2025;Molecular Neurodegeneration2024;Alzheimer’s & Dementia2024;Alzheimer’s Research & Therapy2024)。

②使用大脑神经影像和生物标记物等技术揭示了Aβ病理、tau病理、神经免疫、突触损伤、大脑结构萎缩、皮层糖代谢缺失和认知下降等AD病理的演变规律及作用机制(Biological Psychiatry2021;Annals of Neurology2022, 2023, 2024;Neurology2024;Alzheimer’s & Dementia2025a, b, c, d),为制定AD早期诊疗策略提供科学依据;

③构建PET分子影像精准定量分析活体大脑AD病理新方法,并基于此揭示了AD发生发展中的大脑皮层Aβ斑块(European Journal of Nuclear Medicine and Molecular Imaging2024)和tau缠结(JAMA Neurology2025;Molecular Psychiatry2025;Neurology2023;European Journal of Nuclear Medicine and Molecular Imaging2025;Brain Communications2022)影像病理的空间聚集特征及演变规律,建立了AD影像病理精准分级和早期诊断策略(Neurology2020a;Alzheimer’s Research & Therapy2023);

④以PET分子影像病理指标为标准,进一步阐明了AD发生发展中Aβ病理(Neurology2020b, 2024;Translational Psychiatry2022)、tau病理(Alzheimer’s & Dementia2025e;Alzheimer’s Research & Therapy2020, 2021)以及神经变性(Cell Reports Medicine2025;Alzheimer’s & Dementia2023)等生物标记物的演变规律及相互作用。
