研究领域

应用生物化学、病毒学、人工智能等多学科研究工具,系统研究病毒复制及免疫逃逸机理,为开发基因递送工具奠定基础。包括1)病毒包涵体的发生机制以及体外生化重构,在病毒复制以及同宿主互作中的作用,致病机理及分子诊断。 2)利用前沿生物物理手段,开发对病毒颗粒进行基于形状及内容的分选技术,研究病毒颗粒形状对侵染及致病性的影响,为提升疫苗有效率提供理论基础。3)开发病毒复制小分子抑制剂以及高通量血清学技术。


成果

博士期间,利用核磁共振及细胞药物转运技术,对细菌多重耐药转运蛋白进行了系统的结构-动态-功能研究,为理解跨膜转运以及识别不同类型抗菌药物提供理论基础,为开发新型抗生素提供了思路。

博后及讲师阶段于病理免疫系进行感染免疫方面的生化、结构、细胞、遗传学等多学科研究,对高致病埃博拉病毒、冠状病毒、人类偏肺病毒、合胞病毒的病毒复制机理及免疫逃逸机理进行了系统研究,并筛选了抗病毒小分子抑制剂。疫情期间,探索了新冠病毒N蛋白(核蛋白)作为检测标志物的独特性质,利用核蛋白不同片段,系统研究了血清学检测CTD结构域为特异性及灵敏性最佳标志物,合作开发了高通量血清学方法用于新冠抗体检测,为香港大规模血清学研究探讨mRNA及灭活疫苗的有效性半衰期提供了理论基础,对于新冠疫情期间的公共政策指导做出了重要贡献。

相关工作发表在Annual Review of Virology, Cell, Nature Med., Nature Immunology, iScience, Respirology, ACS Infectious Diseases, Nature Communications, Antiviral Research, mBio 等期刊。埃博拉部分工作收录至最新版病毒学圣经Fields Virology丝状病毒章节。在Trends in Microbiology撰写关于埃博拉病毒复制起始机理的评论一篇。在Annual Review of Virology 撰写了病毒复制形成特殊细胞器的章节。 

吴超博士现为美国病毒协会、微生物协会等行业协会成员,应邀为Nature Immunology, Nature Chemical Biology, mBio, Journal of Medical Virology 等行业期刊审稿。2021年遴选为eLife 期刊Early Career Reviewer, 2022年成为Vaccines 客座编辑及Topical Advisory Board成员,以及NIH COMPASS Scholar。2023年遴选为JBC期刊Early Career Reviewer,获得美国病毒学年会Global Scholar奖项。
